The Clinical Kinase Index (CKI): A user friendly application to prioritize kinases as prospective cancer drug targets

چکیده

Kinases are among the most established druggable proteins with currently over 50 approved kinase inhibitor drugs, of them for cancer. However, these drugs only target a small subset human kinome and many kinases remain “dark” or “understudied” (Essegian et al. 2020 [[1]Essegian D. Khurana R. Stathias V. Schurer S.C. To improve utility evaluate relevance kinases, including their potential novel prospective cancer targets, we developed Clinical Kinase Index (CKI). CKI is an inter interactive web application harmonized datasets extracted from several resources that allows researchers clinicians across solid.

Protein kinases as drug targets in cancer.

Identification of the key roles of protein kinases in signaling pathways leading to development of cancer has caused pharmacological interest to concentrate extensively on targeted therapies as a more specific and effective way for blockade of cancer progression. This review will mainly focus on inhibitors targeting these key components of cellular signaling by employing a technology-based poin...

Phosphoinositide 3-kinases as drug targets in cancer.

The past two years have seen phosphoinositide 3-kinases (PI3Ks) move from being seen as potential targets for chemotherapeutics, to one of them--PI3Kalpha--being generally accepted as validated. A huge amount of work indicated that there was an important role for PI3Ks in tumour progression and, particularly, in the control of proliferation, survival and regulation of the potential oncogene PKB...

Aurora kinases as anticancer drug targets.

The human aurora family of serine-threonine kinases comprises three members, which act in concert with many other proteins to control chromosome assembly and segregation during mitosis. Aurora dysfunction can cause aneuploidy, mitotic arrest, and cell death. Aurora kinases are strongly expressed in a broad range of cancer types. Aurora A expression in tumors is often associated with gene amplif...

Tyrosine kinases as targets for cancer therapy.

Enhanced protein tyrosine kinase (PTK) activity correlates with the development of cancer and other proliferative diseases. The hypothesis that PTK inhibitors may be of value in the treatment of cancer led to the systematic synthesis of selective tyrosine phosphorylation inhibitors (tyrphostins) that show in vitro and in vivo anticancer activity. This review will provide an overview of research...
